Fluorescent Apparatus

 

  • Material: 

 

Fluorescein solution: the Fluorescein compound (46955, Sigma) was dissolved in 0.6% phosphate buffered saline (abbreviated PBS) solution. The molar concentration (C) of the Fluorescein was approximately 8.33 nM. The purpose of using PBS as solvent was to stabilize the pH of the solution at around 7.0.

  • Microplate reader:

 

Modulus™ II Microplate Reader made by Turner BioSystems, Inc. is a high-throughput spectrophotometer designed for microtiter plates. The fluorescence intensity of Fluorescein was measured at excitation wavelength at 525 nm and emission ranged from 510 - 570 nm. The fluorescence plate reader was top-reading.
